Medicinal plants have been used since ancient times to treat various diseases and have supported scientific research in the discovery of new products richly active in pharmacological properties. The objective of this research was to verify the effectiveness of the antibacterial and modulatory activity of the ethanolic extract of the leaves of Ziziphus joazeiro Mart. In this context, phytochemical prospecting, the evaluation of antimicrobial and modulatory activities of antibiotics against multidrug-resistant strains, as well as the non-clinical safety profile by open field test, toxicity, and finally, anxiolytic activity and mechanisms of action of the ethanolic extract obtained from the leaves of this species popularly known as juazeiro. The ethanolic extract was obtained by maceration in ethanol P.A for 72 hours. Antibacterial activity was used to determine the minimum inhibitory concentration against the standard strains of Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. The modulatory action was evaluated using the antibiotics Gentamicin, Norfloxacino and Ampicillin against multi-resistant bacterial species mentioned above. The toxicity and anxiolytic activity and its possible mechanisms were used the zebrafish model. The results obtained in phytochemical prospecting suggest the presence of the metabolic components of flobabenic tannins, leucoanthocyanidins, flavonoids, flavonones, catechins, triterpenoidal alkaloids. In the open field test, a decrease in motor capacity was observed when compared to Diazepam, thus suggesting that the extract may have anxiolytic activity. The toxicity test associated with the open field showed a (LD50 ˃ 500
mg / kg), being preclinically safe in this model. The extract has no clinically relevant antibacterial activity, since a high minimum inhibitory concentration was obtained. However, it was effective in modulating the action of antibiotics against multidrug-resistant strains of Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. In the anxiety model, EtFoZj at the following concentrations (10, 100 or 500 mg / kg; ip) had a similar anxiolytic effect (p> 0.05) similar to the effect of Diazepam (500 mg / kg; ip; p <0 , 01 vs. Naive or vehicle). In view of the demonstrated anxiolytic activity, the possible mechanisms of action for these activities were evaluated. Involvement with the following systems was tested: Gabaergic and serotonergic (5-HT2A, 5-HT1 and 5-HT2A / 2C, 5-HT3A / 3B). The extract is possibly acting in the same way as Diazepam because, in the gabaergic test, it suffered an antagonistic action from Flumazenil. Therefore, this study adds new evidence about the potential of Ziziphus joazeiro for the development of herbal medicines with anxiolytic and modulatory properties.
